Screening and serodiagnosis potential of proteins related with multi-drug resistant Mycobacterium tuberculosis |

Abstract Objective To explore the differentially expressed proteins in multi-drug resistant(MDR)strains of Mycobacterium tuberculosis(TB)and their potential for serodiagnosis. Methods Differentially expressed proteins were detected from clinical isolates of MDR TB by protomic technology,compared with standard strain H37Rv. The interested protein spots were identified by mass spectrometry and database searching. The target antigen NuoB was cloned and purified from Escherichia coli. Its IgG levels induced from MDR TB patients,drug-sensitive TB patients and healthy people were detected by enzyme-linked immuno sorbent assay(ELISA).Results Totally 11 differentially expressed proteins in MDR TB strains were detected and 8 proteins were identified by mass spectrometry. NuoB showed both higher expression level in clinical MDR strains and stronger B cell epitope. The serum IgG positive rate of purified NuoB was 44% in MDR TB patients,which was higher than that in drug-sensitive TB patients(15.62%)and healthy subjects(10.00%,P<0.05).Conclusion NuoB protein can be a useful diagnostic marker in MDR TB.
