Application of UPLC-FLD technology to detection of fluorescent whitening agents in edible fungi |

Abstract Objective To apply ultra performance liquid chromatography-fluorescence detection(UPLC-FLD)to detecting various fluorescent whitening agents in edible fungi. Methods We obtained the comminuted edible fungi samples by water bath heating and ultrasonic extraction. We detected the content of fluorescent whitening agents in 11 edible fungi edible by UPLC-FLD.The optimal chromatographic conditions were as follows:ACQUITY UPLC BEH C18 chromatographic column(1.7 μm,2.1 mm× 100 mm);acetonitrile / methanol mixed solution and methanol aqueous solution containing 20 mmol four butyl ammonium bromide for gradient elution. Results Within the concentration ranged from 25 to 1 000 ng/mL,the linear relationship was good because the correlation coefficient was more than 0.99. The detection limit of the 11 fluorescent brighteners was 0.20-0.29 mg/kg and the quantitative limit was 0.67-0.97 mg/kg. In the concentration of 1.0,5.0 and 10.0 mg/kg,the recovery rate of the addition standard was 81.6%-103.5%,the relative standard deviation(RSD)of the samples was less than 10%. Conclusion This method has high sensitivity,good repeatability,ideal recovery rate and linearity for detection of fluorescent brighteners in edible fungi.
