乳铁蛋白和2'-岩藻糖基乳糖复合粉对小鼠免疫功能的调节作用

摘要 目的 探讨乳铁蛋白和2'-岩藻糖基乳糖(2'-FL)复合粉对小鼠免疫功能的调节作用。方法 40只雌性ICR小鼠随机分为对照组和低、中、高3个剂量组(0.125、0.250、0.750 g/kg体重),每组各10只。低、中、高剂量组小鼠每日经口灌胃给予10 mL/kg体重的乳铁蛋白和2'-FL复合粉,对照组小鼠给予等体积去离子水,连续30 d。采用小鼠脾淋巴细胞转化实验和迟发型变态反应实验分别检测光密度差值和左后足跖部厚度差值;采用血清溶血素检测和抗体生成细胞检测实验分别测定抗体积数和溶血空斑数;采用小鼠腹腔巨噬细胞吞噬鸡红细胞实验检测巨噬细胞数,计算吞噬率和吞噬指数;采用自然杀伤细胞活性实验检测光密度值,计算NK细胞活性率。结果 与对照组相比,高剂量组小鼠光密度差值和左后足跖部厚度差值升高,中、高剂量组小鼠腹腔巨噬细胞吞噬鸡红细胞的吞噬率和中剂量组小鼠腹腔巨噬细胞吞噬鸡红细胞的吞噬指数升高(均P<0.05)。4组小鼠抗体积数、溶血空斑数和NK细胞活性率比较,差异无统计学意义(均P>0.05)。结论 乳铁蛋白和2'-FL复合粉可通过增强细胞免疫功能和单核-巨噬细胞功能发挥免疫调节作用。

AbstractObjective To explore the immunomodulatory effect of lactoferrin and 2'-fucosyl-lactose (2'-FL) compound powder on immune function in mice. Methods Forty female ICR mice were randomly divided into a control group and three dose groups (0.125, 0.250, and 0.750 g/kg body weight), with 10 mice in each group. The three dose groups were intragastrically administered the compound powder solution at 10 mL/kg body weight daily, while the control group received an equal volume of deionized water for 30 consecutive days. The splenic lymphocyte transformation test and delayed-type hypersensitivity test were used to measure the optical density difference and the left hind paw thickness difference, respectively. Serum hemolysin assay and antibody-forming cell assay were performed to determine the antibody volume and the number of hemolytic plaques. The peritoneal macrophage phagocytosis assay with chicken red blood cells was conducted to calculate the phagocytosis rate and phagocytic index. The natural killer (NK) cell activity assay was used to measure optical density and calculate NK cell activity. Results Compared with the control group, the optical density difference and left hind paw thickness difference were significantly increased in the high-dose group, the peritoneal macrophage phagocytosis rate of chicken red blood cells was significantly increased in the medium-dose and high-dose groups, awhile the phagocytic index was significantly elevated in the medium-dose group (all P<0.05). There were no significant differences in antibody volume, hemolytic plaque number or NK cell activity among the four groups (all P>0.05). Conclusion Lactoferrin and 2'-FL compound powder exerts an immunomodulatory effect by enhancing cellular immunity and monocyte-macrophage function.
